Development of highly efficient whole-cell catalysts of cis-epoxysuccinic acid hydrolase by surface display

Abstract Bacterial cis -epoxysuccinic acid hydrolases (CESHs) are intracellular enzymes used in the industrial production of enantiomeric tartaric acids. The mainly as whole-cell catalysts because low stability purified CESHs. However, cell permeability is major drawback catalyst. To overcome this problem, we developed using various surface display systems for CESH[L] which produces L(+)-tartaric acid. Considering that efficiency depends on both carrier and passenger, screened five different anchoring motifs Escherichia coli . Display efficiencies significantly among these InaPbN-CESH[L] system has highest enzymatic activity. Conditions were optimized a maturation step was discovered can increase activity several times. After optimization, total higher than lysate an overexpression system, indicating very high level. Furthermore, biocatalyst exhibited good storage at 4 °C considerable reusability.
